Abstract

Bagasse can be used as a basic ingredient in the manufacture of bio-ethanol and cellulose in bagasse first should be released from lignin by using pretreatment method. Pretreatment was done by using a mixture of 2% NaOH with NH4OH concentrations that varied from 2% to 10%. The solids ratio variation (bagasse): liquid (NaOH-NH4OH) pretreatment was 1:10, 1:15, and 1:20. Pretreatment condition which provided the optimum sample weight reduction was at concentration of 2% NaOH and 2% NH4OH with a ratio of 1:20. The results of bagasse saccharification using cellulase enzyme crude extract of Trichoderma viride strain T1 sk against CMC pure substrates showed enzyme activity of 0,188 units. Saccharification was also done to the variation of the amount of bagasse substrate 0,1g to 1 g and the variations of the time of saccharification from 30 min to 120 min. The maximum glucose concentration was 1528.57 µg / mL which obtained at 0.6 g substrate of bagasse with a long of saccharification 60 minutes. Bio-ethanol obtained by the method of SSF for 84 hours with 30 mL inoculum of Saccharomyces cerevisiae and 10 mL of crude extract of the enzyme cellulase was determined by using gas chromatography, is equal to 0,4278 mL.
