Abstract

Determination of BAC-end terminal copy number is a crucial step during contig assembling. This can be as direct probe for clones ordering via ordinary hybridization or indirect use for searching new clones to fill the existing gaps among the available contigs. In this paper we compare the estimation results obtained from PCR technique and southern-hybridization method. The results indicated differences between both methods, where PCR-based technique showed single- and low-copy fragments almost two times more than southern hybridization-based method. However the PCR-based technique indicated consistency with the successfulness of PCR-based marker development. Besides, PCR based technique provides acceleration and safety in copy number determination. Key words: Jumlah salinan, ujung-ujung insersi klon BAC, PCR
