Abstract

The experiment on the growth and development of bitter nuts shoot tip in-vitro at different concentration of NAA and BAP in MS media was carried out at Plant Tissue Culture Laboratory Department of Agronomy, Faculty of Agriculture, Andalas University from September 2002 to January 2003. The objective of the experiment was to obtain the suitable concentration of NAA and BAP for the growth and development of bitter nuts shoot tip by in-vitro culture. Two factor treatments were arranged in Completely Randomized Design. The first factor was NAA with four levels i.e.: 0 ppm; 1 ppm; 2 ppm; and 4 ppm. And the second factor was BAP with four levels i.e.: 0 ppm; 1 ppm; 2 ppm and 4 ppm and there replication. Data were analyzed by using F test, then continued with Duncan’s New Multiple Range Test (DNMRT) at 5 % level. The result indicated that MS media without addition of NAA and BAP, could promote growth and development of bitter nuts shoot tip. Increasing NAA concentration to 4 ppm gave a high percentage of callus growth. The effect of addition of NAA and BAP showed a dynamic influence to explant growth.
