Abstract
An experiment was aimed at determining suitable isoenzymes as genetic marker in characterising three genotypes of gambier plants, i.e. Udang, Cubadak, and Riau. These genotypes were compared to other six genotypes of gambier of the collection of Faculty of Agriculture, Andalas University. Gambier leaves were subjected to analytical method following a modified-technique of Wendel dan Weeden (1989). Isoenzymes analysis was carried out as the following stages: preparation of buffer, preparation of starch gel, enzyme extraction, electrophoresis, and staining. Results demonstrate: 1) isoenzyme estherase, proxidase, and ACP can be used to determine genetic relatedness and grouping in gambier plants; 2) gene controlling isoenzyme estherase, proxidase, and ACP were identified at two loci, and 3) there were three groups of gambier genotype found according to the relatedness analysis of isoenzyme estherase at 36.75% similarities; isoenzyme peroxidase at 29.29% similarities shown two groups of gambier genotypes; and isoenzyme ACP at 36.75% similarities revealed three groups of gambier genotypes.
