Abstract

An experiment was carried out at the plant tissue culture Laboratory of Department of Agronomy, Faculty ttf Agri- culture, Andalas University, Padang from September to December 1999. The objective of the experiment was to obtain the best combination af 2,4-D and kinetin to in- crease callus induction, and combination of NAA, kinetin and BAP to increase callus generation of gambir in vifto. Two factor treatments lvere arranged in Completely Ran- domized Design (CRD) with 4 replications. The first factor was concentration of 2,4-D i.e:0,0; 0,5; and 1,0 ppm and the second factor was concentration of kinetin i.e :0,0; 0,5; 1,0; and 1,5 ppm. For stage callus regeneration, callus formed on stage callus induction were cultured on combi- nation concentration of 0,5 ppm NAA + 0,1 ppm kinetin + 3,0 ppm BAP. Observations were on percentage of browuing, percentage of callus, percentage of live, and percentage of shootlet. Result indicated that the interac- tion of concentration of 0,5 ppm, 2,4-D and 0,5 ppm ki- netin was the best in callus induction. For stage callus regeneration formed shootleto rootlet, and planlet on com- bination concentration of 0,5 ppm NAA + 0,1 ppm kinetin + 3,0 ppm BAP.
