Abstract

The Aspergillus niger have been mutated, screened and applied for glucose oxidase production. The A. niger was mutated by irradiation of its suspension spore with gamma ray at doses 3,3,5, 4,0 and 4,5 kGray, and then spores were spread over the potato dextrose agar (PDA) medium. The survival spores, that grows after two days incubation were isolated and considered as mutant. There were 63,18, 4 and 0 (zero) mutants isolated from irradiated spores at dose 3,3,5,4, and 4,5 respectively. The mutants were screened for the production of glucose oxidase on screening medium containing K3[Fe(CN)6] and FeCl3 as chromogen. It was detected 25 out of 85 mutants that excreted GOD to the medium. It were detected that 9 out of 25 mutants developed higher diameter of blue area around their colonies (produced higher GOD) then developed by the wild type. Next, the nine mutants were tested their ability to produce GOD on fermentation. Four of the nirre mutants produced higher GOD than by wild type on fermentation broth. They are A. niger mutant B10, C31, C32 and C33 which produced GOD 1,38, 1,54, 1,89 and 1,3 time higher respectively then by wild type. Key Word; Aspergillus niger, mutation, glucose oxidase.
