Abstract

Biochemical selection have been very useful to select high alkaloid ciones in Catharantr roseus plant. Clonal variation have also been created after gamma-irradiation. Several doses have been used to produce clonal variation. The most effective doses used to perform better clonal variation was about 15 to 20 krad. The frequency of resistant variants was l0-1. The resistant variants r,vere germinate and grorvn better than the wild qvpe. Biochemical selection used to the resistant variants was analog substrate of arnino acid tryptophan (5-Metyltryptophan) or (5-MT). Several concentration of 5-MT subsequentl;- ( 0; 0.05; 0.1; 0.15;0.2 mM) were treated to the resistant variants. The highest concentration to select biochemical mutants was 0.2 mM. We have success to get (F2) seeds from the expected mutant. The seeds from selected mutant(M2) are bigger when comparing to the wild type and growth better on medium containing (5-MT). We expected higher tryptophan over-producing mutants as a precursor of indole alkaloid. Further experiments continue to do invitro culture in order to develop embryonic callus from leaf tip and leaf base. Several manipulations of auxin and cytokinin have been used to differentiate the callus formation. Modified MS medium r.vith kinetin and cytokinin ( l0: I ) can induce globular embryo like structure. Dragendrof alkaloid reagen were used to determine hi_eh alkaloid clones from the expected mutant. TLC analysis from callus mutant shows 3 clear bands with subsequent Rf about 0.22,0.41,0.58 rvhile control shor,vs trvo smearing bands at0.2t and 0.52.
