Abstract
Experiment to invistigate (Plant Propagation Agarwood ((Aquilaria malaccensi Lamk) by Effort Conservation Plasma Nutfah to in Vitro) was done in February to November 2009 at tissue cultur laboratory of Agriculture Faculty, University Andalas Padang and at tissue cultur laboratory of Mapeni Padang. The experiment consist of tri tree stages, The first stage about give eksplant and Medium for induction callus, seven (9) level consist of : 1).Shootlet+Medium MS; 2). Shootlet+ Medium WPM; 3). Shootlet+Medium B5; 4). Ibu tulang daun +Medium MS; 5). Ibu tulang daun +Medium WPM; 6). Ibu tulang daun+Media B5; 7) Petiole+Medium MS; 8). Petiole + Medium WPM and 9). petiole+Mediium B5. The second stage about give combination concentration 2,4-D and BAP for regeneration callus,seven (9) level consist of : 1). 0,00 ppm 2,4-D + 0,00 ppm BAP; 2) 2,50 ppm 2,4-D + 0,00 ppm BAP; 3). 2,50 ppm 2,4-D + 1,50 ppm BAP; 4). 2,50 ppm 2,4-D + 3,00 ppm BAP(K3); 5).5,00 ppm 2,4-D + 0,00 ppm BAP (K4); 6).5,00 ppm 2,4-D + 1,50 ppm BAP; 7). 5,00 ppm 2,4-D + 3,00 ppm BAP; 8) 7,50 ppm 2,4-D + 0,00 ppm BAP dan 9). 7,50 ppm 2,4-D + 1,50 ppm BAP. The third stage about give combination concentration NAA,BAP and Kinetin for regeneration callus,seven (10) level consist of : 10.0,00 ppm NAA+0,00 ppm Bap + 0,00ppm Kinetin; 2)..0,50 ppm NAA + 1,50 ppm BAP+ 0,10 ppm Kinetin; 3).0,50 ppm NAA + 3,00 ppm BAP + 0,10 ppm Kinetin ; 4). 0,50 ppm NAA + 4,50 ppm BAP + 0,10 ppm Kinetin; 5). 1,00 ppm NAA + 1,50 ppm BAP + 0,10 ppm Kinetin; 6). 1,00 ppm NAA + 3,00 ppm BAP + 0,10 ppm Kinetin ; 7). 1,00 ppm NAA + 4,50 ppm BAP + 0,10 ppm Kinetin ;8). 1,50 ppm NAA + 1,50 ppm BAP + 0,10 ppm Kinetin; 9). 1,50 ppm NAA + 3,00 ppm BAP + 0,10 ppm Kinetin and 10). 1,50 ppm NAA + 4,50 ppm BAP + 0,10 ppm Kinetin (R9),The experiment used Completely Ramdomized Design (CRD) with four (replication). Explant Ibu tulang daun and Medium MS were superior result than any another treatment the support growth to get explants life, callus percentage callus, callus formation, less browning for first stage. Combination Concentration 2,50 ppm 2,4-D + 3,00 ppm BAP) were supperior result than any another treatment the support growth to get explant life, percentage regeneration callus formatted shootlet for secallus formation , for second stage. Experiment third stage can not observated because Eahrthquake. Key Words : Explant, Induction callus,Regeneration callus, Callus, Shootlet, 2,4-D, Auxin NAA, Cytokinin BAP, Kinetin and in vitro.
