Abstract

Experiment to invistigate by Effort to improve somaclone variation plant Agarwood (Aquilaria malaccensi L ) to in vitro was done in Mei to December 2008 at tissue cultur laboratory of Agriculture Faculty, University Andalas Padang and Institute Agriculture Bogor, Bogor. The experiment consist of two serries, The first serries about give concentration 2,4-D for induction callus, seven (7) level consist of : 0,00 ppm (A1); 5,00 ppm (A1); 10,00 ppm (A2); 15,00 ppm (A3); 20,00 ppm (A4); 25,00 ppm (A5); dan 30,00 ppm (A6) The second serries about give combination concentration NAA and BAP for regeneration callus,seven (5) level consist of : 0,00 ppm NAA + 0,00 ppm BAP (K0); 5,00 ppm NAA + 0,00 ppm BAP(K1); 5,00 ppm NAA + 5,00 ppm BAP(K2); 5,00 ppm NAA + 10,00 ppm BAP(K3); 5,00 ppm NAA +15,00 ppm BAP(K4); 1,0 ppm NAA + 0,10 ppm Kinetin + 3,50 ppm BAP(K5); dan 1,0 ppm NAA + 0,10 ppm Kinetin + 5,25 ppm BAP(K6) The experiment used Completely Ramdomized Design (CRD) with four (replication). Concentration 15,00 ppm 2,4-D (A3) were supperior result than any another treatment the support growth to get explant life, callus formation , and less browning. Combination concentration 1,00 ppm NAA + 3,50 ppm BAP (K5) were supperior result than any another treatment the support growth to get callus life, and regeneration callus formed shootlet. Key Words : Explant, Induction callus,Regeneration callus, Callus, 2,4-D, Auxin NAA, Cytokinin, and in vitro.
