Abstract
Experiment induction callus mangosteen (Garcinia mangostana L.) by in vitro culture was done in August to December 1997 . The objective of this experiment was to get medium composition that exactly to support induction callus mangosteen by in vitro culture and to get plantlet. The experiment used Corn-pletely Randomized Design (CRD) with Five (5) replication. Stage induction callus on Woody Plant Medium (WPM) with combination concentration Auxin and Cy- tokinin plant growth regulator four (4) level treatment, consist of : l), 2.00 ppm 2.4-D + 2.00 ppm Ki-netin (Kr); 2), 1.00 ppm 2.4-D + 1.00 ppm NAA + 1.00 ppm Kinetin (Kj; 3), L.75 ppm Kinetin + 0,25ppm NAA (Kr); 4), 135 ppm Kinetin + 0.50 ppm NAA (K4). Combination Concentration 2.00 ppm 2.4-D + 2.00 ppm Kinetin (Kr) were superior result than any another treatment the support induction callus. Percentage explant browning and contamination were lower result on combination concentration 2.00 ppm 2.4-D + 2.00 ppm Kinetin(Kr) than any another freatment. Percentage explant life and callus formation were higher result on combination concentration 2.00 ppm 2.4-D + 2.00 ppm Kinetin (K1) than any another treatment. The result of this study show that effect medium composition WPM + 1,75 ppm BAP + 0,50 ppm NAA and effect medium composition WPM + 3,00 ppm BAP + 0,01 ppm Kinetin + 0,50 ppm NAA with plant organ cotil ring were supperior result than any other treatments on the inisition and proliferation of mangosteen in vitro culture.
