Abstract

Quercetin, a chemical cousin of the glycoside rutin from Manihot esculenta Crantz , is appears to have many biologically active. Preformulation studies are usually associated for a new drug substances, would include intrinsic solubility, ionization constants (pKa) and stability indicating assay. The determination of solubility and ionization constants were calculated by phase solubility and potentiometric titration methods, respectively. The research was done by intrinsic solubiliy of quercetin is 3,2 ± 0,07 μg/mL in HCl 0,1 N. The complexation of quercetin by β-cyclodextrin indicated an AL-type diagram with inclusion complex of 1:1 molar ratio and stability constant K is 403,58 M-1. Enhancement of solubility was 9-fold with the quercetin, but not efective to liquid and oral phamaceutical applications. The pKa values are pKT1 = 7,30 ± 0,12 dan pKT2 = 8,63 ± 0,18. Method of analysis determined by HPLC with methanol-water-phosporic acid as mobile phase are not stability indicating assay
