Abstract

Cancer is the second leading cause of death after cardiovascular diseases in the world. Multiple Myeloma has a 10% prevalence of all plasma cell cancer cases. Curcumin is a compound isolated from the rhizome of turmeric (Curcuma longa L.) and has been proven to hare an activity as anticancer. One of the new curcumins analogues compound was 2,5-bis(2'-pyridilidine)cyclopentanone. This study was aimed at the synthesis and determination of cytotoxic activity of 2,5-bis(2'-pyridilidine)cyclopentanone against the Myeloma. Synthesis of 2,5-bis(2’- pyridylidine)cyclopentanon was performed according to carbonil condensation reaction between cyclopentanon and pyridtne-2-catbaldehyde with potasium hydroxide as catalyst. The purity of the synthesized product was analyzed by measuring melting point, Thin Layer Chromatography (TLC), and Gas Chromatography(GC). Then the structure of the compound was identified by using spectroscopic methods (UV-Vis, IR, H1 -NMR, and Vero Mass spectroscopic). MTT Test was used as the method of the cytotoxic activity test in the Myeloma and Vero cells. The series of concentration used in the Myeloma cell for the 2,5-bis(2'-pyridilidine)cyclopentanone were 200 µM; 100 µM, 50 µpM; 25 µM;12,5 µM;6,25 µM. In the Vero cell, the series of concentration used for 2,5-bis(2,-pyridilidine)cyclopentanone were 400 µM; 200 µM; 100 µM, 50 µM. The absorbance of the cell was measured at 550 nm wavelength using the ELISA reader. IC50 was used as a cytotoxicify parameter was obtained from probit analysis. Identification by UV-Visible, IR, 1H-NMR and MS showed that the synthesized product was 2.5-bis-(2'-pyidilidine)cyclopentanon. The TLC test identified one spot and GC showed 92.03% purity. The 2,5-bis(2'-pyridilidine)cyclopentanone had cytotoxic effect to Mye loma cel l. Keywords: synthesis ,2,5-bis(2'-pyridilidine)cyclopentanone, curcumin, Myeloma cell, Vero cell, Cytotoxic
