Abstract

The rhizosphere is an area where the intense interaction between root and soil microorganisms occurs. The total population and biodiversity of soil microorganism vary among the plants and highly be influenced by root exudate. Indole Acetic Acid (IAA) producing rhizobacteria in this research were isolated from Tithonia diversifolia (a plant that has a potential to use as a green manure) rhizosphere. Tithonia diversifolia withstands well in acid soil that has poor nutrient. This experiment was conducted to isolate the rhizobacteria capable producing IAA and characterize the capability of the isolate in producing IAA to several media conditions i.e the several degrees of acidity, temperatures, triptophan and C, N and P sources in media. The isolation had conducted in King’B agar. The isolates had shown the ability to produce IAA were purified by using the same media before being characterized. The pH, the temperatures of culture incubation, the addition of triptophan, and C, N and P sources into media were used in characterization. Results revealed that IAA producing isolates obtained could be distinguished into two groups according to their colony characteristics. We obtained two isolates belonging to milky white and platy round colony (Ti.1a, Ti.3a) and two isolates belonging to transparent and platy round colony (Ti.1b, Ti.2b). The Ti.2b isolate exhibited as an acid tolerant and could grow in pH 4.0 agar media. The presence L-triptophan in broth medium culture could enhance IAA production of four isolates. If these isolates are grown in soil extract broth culture, the addition of C,N and P sources are needed for IAA production. The optimum temperature for the growth of four isolates was 300C. According to IAA production ability we can state that although the four isolates have the similarity in colony chararacteristics but they most probably come from different species.
